The PNC-27 Peptide is a cancer-targeting peptide that binds to cancer cell membranes via the HDM-2 receptor binding, and disrupts the cancer cell’s membrane, thereby leading to the cancer cell’s death. PNC-27 was originally conceptualized and developed in 2000 by Drs. Matthew Pincus and Joseph Michl at the SUNY Downstate Medical Center in New York.
PNC-27 is a synthetic peptide derived from the tumor suppressor protein p53, one of the most important regulators of genomic stability and cellular death. Rather than acting as a transcription factor like native p53, PNC-27 operates through a distinct and unconventional mechanism: direct interaction with cancer cell membranes, disrupting these membranes by creating perforations and ultimately leading to the cancer cell death. More specifically, binding with this protein appears to perforate the membrane of cancer cells forming holes and causing membranolysis. This allows for the influx of ions and the efflux of cellular contents, ultimately leading to cell death.
